342,43 Aloxistatin ist ein irreversibler und membrandurchlässiger Cystein-Protease-Inhibitor.
Biologische Aktivität Aloxistatin kann in die intakten Blutplättchen eindringen und die Proteolyse durch Hemmung von Calpain hemmen. Aloxistatin stumpft die durch Parathormon (PTH) induzierte Zellproliferation ab und hemmt die Differenzierung von Osteoblasten in vitro. Aloxistatin (100 mg/kg, po) hemmt stark die Cathepsin-B&L-Aktivitäten in Skelettmuskulatur, Herz und Leber von Hamstern. Bei Ratten mit Rückenmarksverletzung (SCI) bietet Aloxistatin Neuroprotektion bei SCI-Läsion und Penumbra. Aloxistatin reduziert Amyloid-β im Gehirn und verbessert Gedächtnisdefizite in Alzheimer-Tiermodellen durch Hemmung der Cathepsin-B-Aktivität.
