Biologische Aktivität ARRY-380 ist ein oral bioverfügbarer, potenter, selektiver, niedermolekularer Tyrosinkinase-HER2- und p95-HER2-Inhibitor mit einer IC50 von 8 bzw. 7 nM. Allerdings hemmt ARRY-380 EGFR mit einem IC50 von 4 μM. ARRY-380 ist etwa 500-fach selektiv für HER2 gegenüber EGFR. ARRY-380 produziert signifikante Antitumoraktivitäten in mehreren HER2-abhängigen Tumor-Xenograft-Modellen und erzeugt additive Aktivität in Kombination mit Standard-of-Care-Wirkstoffen.
