Nizatidin, ein selektiver Histamin-H2-Rezeptor-Antagonist, ist a potenter Inhibitor der Magensäuresekretion, mit IC50 von 0,9 nM. Nizatidin hemmt auch reversibel die Acetylcholinesterase (AChE) mit einer IC50 von 6,7 µM, und die Hemmung ist mit einem Ki-Wert von 7,4 µM nicht kompetitiv. Nizatidin zeigt eine maximale Hemmung der Magensäure bei Ratten innerhalb der ersten Stunde nach Verabreichung des Arzneimittels mit einer EC50 von 1,383 μmol/kg. Nizatidin (0,3-3 mg/kg, iv) erhöht dosisabhängig den motorischen Index der gastrointestinalen (GI) Motilität signifikant. Nizatidin hemmt die Magensäuresekretion mit ED50 und ED90 von 0,18 bzw. 3,22 mg/kg bei Hunden bzw. 2,94 bzw. 19,6 mg/kg bei Ratten.
