Molekulargewicht:
Etheno-ATP fluorescent assay
|
Human Arp2/3 complex (0.7 μM) is incubated with 2.5 μM etheno-ATP, 0.2 M acylamide (used as a etheno-ATP fluorescence quencher) in 2mM Trs-HCl pH 8.0, 50 mM KCl, 0.05 mM EGTA 0.8 mM MgCl2, 0 and 0.5 mM DTT. Ethano-ATP emission spectrums are acquired upon excitation at 340 nm using spectrofluorometer FluoroMax. Inhibition of Arp2/3 complex by 100 μM CK-636 is performed in the presence or absence of 2.8 μM WASp-VCA.
